Experiment Description - Organism: Homo sapiens - Cell line: RPMI-8226 multiple myeloma cell line - Library used: Brunello Human CRISPR Knockout Pooled Library (Addgene #73179) - Cas9 system: Cells stably expressing SpCas9 - Selection marker: Endogenous EGFP-tagged MYC - Experimental design: Cells were transduced with the pooled sgRNA library, selected, expanded, and viable cells were sorted into MYC-GFP-high, -intermediate, and -low expression fractions based on GFP fluorescence to enrich for repressors and activators of MYC expression, respectively. - Screening strategy: Genome-wide loss-of-function screen; comparisons were made between sorted high vs. intermediate and low vs. intermediate MYC-GFP populations. ### 2.
